My Psoriasis Revolution Review

Highly Recommended

I usually find books written on this category hard to understand and full of jargon. But the writer was capable of presenting advanced techniques in an extremely easy to understand language.

This book served its purpose to the maximum level. I am glad that I purchased it. If you are interested in this field, this is a must have.

The Psoriasis Strategy

Psoriasis is commonly referred to as a skin condition that quickness up the life cycle of skin cells. It is a condition that causes cells to build up rapidly on the surface of the skin. The extra skin cells form white patches known as scales and red patches, which are itchy and sometimes painful. Psoriasis is not a skin problem per se, but an inflammation problem which is connected to the immune system. The inflammation of the skin is what leads to a flaky, itchy, red skin condition that people living with psoriasis are used to. People suffering from this condition experience discomfort and low self-esteem the entire time. They have problems communicating, getting into intimate relationships, and being in public because all the time, they meet new people who will notice their condition. The only time psoriasis sufferers get comfortable is when they are indoors or in forums where they are all victims of the same situation. This condition lasts longer in some patients as compared to others. Medications, ointment, among other treatments, have been offered for psoriasis patients. The medicines seem to work and relieve pain for a shorter period then the side effects become even worse. The solution is simple and is known to many people. Continue reading...

Modulation of Lymphocyte Homing for Therapeutic Purposes

Some genetic defects in adhesion mechanisms result in variable degrees ofim-munodeficiency, for example, leukocyte adhesion deficiency (LAD)-1 (lack of functional 02 integrins) (Kishimoto et al. 1987), LAD-2 (deficiency in selectin ligands) (Etzionietal. 1992), or LAD-3 (defects in integrin activation) (Kinashi et al. 2004). These paradigmatic experiments of nature imply that the manipulation of leukocyte adhesion homing could be a useful immunotherapeutic tool. In fact, a mAb against the integrin LFA-1 (efalizumab) is being used in the clinic for the treatment of psoriasis (Lebwohl et al. 2003). Anti-LFA may also be effective in the treatment of refractive cutaneous GVHD (Stoppa et al. 1991) and possibly other inflammatory immune-mediated diseases. However, blocking either LFA-1 or one of its ligand ICAM-1 has not been successful in stroke, myocardial infarction, and traumatic shock (Schreiber et al. 2001 Yacyshyn et al. 2002 Yonekawa and Harlan 2005). On the other hand, because...

PTK Receptors and Disease

Given the importance of PTK receptors in the control of cell proliferation and migration, it is not surprising that over-activity of PTK receptors occurs in cancer and other diseases that involve excess cell proliferation, such as inflammatory and fibrotic conditions and psoriasis. About half of the PTK receptors are implicated in various human malignancies (Fig. 1) 2 . Often, the receptors are constitutively activated by amplification or mutational events. Several mutations of PTK receptors cause constitutive dimerization (1) by mutations affecting disulfide bonding in the extracellular parts of the receptors, thus causing the formation of covalent dimers, mutations of other residues in the transmembrane, or jux-tamembrane domains that promote dimerization or (2) by formation of fusion proteins between the kinase domains of the receptors and proteins that normally occur as dimers or oligomers. The end result is a constitutively active kinase that drives cell growth.

The Sympatho AdrenalMedullary System

In hypertension, a role for elevated norepinephrine levels has been established (DeQuattro and Feng, 2002). Epinephrine may furthermore interact with immune functions. In HIV, stress may be involved in disease progression partly due to epinephrine effects on HIV-infected leucocytes (Cole, 2008). In psoriasis and atopic dermatitis, a hyperre-sponsive plasma catecholamine response to psychosocial stress may be involved in dysreg-ulations in Th1 Th2 mediated immune states (Buske-Kirschbaum et al, 2006). The sympatho-adrenal-medullary system may also play a role in the pathogenesis of obesity. Epinephrine

Spatiotemporal patterns of common skin diseases

Despite the absence of systematic data there are important secular and geographical differences in disease burden that (to this author) seem robust. Atopic dermatitis incidence appears to have increased by a factor of two or more in the last three decades in many developed countries (Williams, 1997). Cancers of the skin in many world populations (chiefly those with pale skin) have also increased two to three-fold over a similar time course (Marks, 1995). By contrast psoriasis incidence seems to have stayed constant. For other

Atopic dermatitis OMIM 603165

Sib and other family studies have identified a number of loci, some of which show overlap with those implicated in psoriasis. The genes underpinning the linkage are not known. Special mention should be made of the gene underlying Netherton's syndrome (OMIM 256500, SPINK5).

Clinical Manifestations

NME is the hallmark finding in glucagonoma syndrome. It is characterized by a polymorphous eruption that most commonly presents as scaly, erythematous papules and plaques with superficial erosions. The lesions typically are in a perioral distribution on the face (Fig. 9), but also involve the perineum, lower abdomen, thighs, buttocks, and less commonly the distal extremities. NME has been reported to be the presenting complaint in approximately two-thirds of patients with this tumor, but there are only rare cases of glucagonoma without NME ever occurring. Stomatitis, glossitis, dystrophic nails, and alopecia can also be seen. The eruption is frequently misdiagnosed as seborrheic dermatitis or intertrigo and can also resemble pemphigus foliaceus, acrodermatitis enteropathica, chronic mucocutaneous candidiasis, or psoriasis.

Scope of chapter

I will take a selective approach dealing with the most common diseases acne, dermatitis, psoriasis and skin cancer account for over half the dermatology cases seen in primary care and around 80 of the workload of a secondary care dermatologist (Harris et al., 1990). What (little) we know of the genetics of acne, atopic dermatitis and psoriasis will be summarized briefly, and the role of genetic factors in skin cancer dealt with in more detail.
